Product Description

M9 Minimal Salts are a commonly used base for preparing a defined synthetic medium for the growth of most E. coli host cells. The powder is reconstituted in sterile purified water, (or water with melted purified agar added.) Additional nutrients, such as glucose or other carbon source, casamino acids, vitamins, or antibiotics are added when the solution cools and before plates are poured or media is inoculated with bacteria.

M9 Minimal medium salts are suitable for the preparation of M9 Minimal Medium for use in propagation of E. coli and plasmid amplification. The resulting M9 medium can be used for maintaining positive selection pressure on plasmids coding for genes to produce essential substances such as amino acids and vitamins and for maintenance of F’-containing bacteria strains for M13 propagation.

Preparation Method Media Preparation:Reagents are tested and chosen for their ability to promote optimum growth of E. coli when combined in M9 Minimal Medium Salt formulations. Chosen reagents are combined in the proper proportions according to the formulation listed in the Certificate of Analysis and thoroughly blended for a uniform distribution.

Product Overview Parts A and B are initially dissolved in water, autoclaved, and thereafter serve as 50x concentrated stock solutions for making M9 Growth Media. To make M9 Growth Media, 2% parts A and B are added to sterile water (or water plus melted, purified agar). Additional nutrients, such as glucose, casamino acids, vitamins, or antibiotics are added when the solution cools and before plates are poured or media is inoculated with bacteria.

Sterilization of Solutions A 45 g sample of M9 Minimal Salts Part A was dissolved in water to 100 mL, the solution was autoclaved to make a sterile 50 X concentrate. A 8.4 g sample of M9 Minimal Salts Part B was dissolved in water to 10 mL, the solution was autoclaved to make a sterile 50 X concentrate.
